What is Dayvigo 5 mg?
Dayvigo is a film-coated tablet whose active ingredient is lemborexant, 5 mg per tablet, made by Eisai Pharmaceuticals India Private Limited in India. A box holds 28 tablets (2 strips of 14). How to use Dayvigo 5 mg
Dose according to the package insert
Once a day
5 mg no more than once per night, taken immediately before going to bed, with at least 7 hours left before the planned time of waking. Depending on response and tolerability, a doctor may raise it to a maximum of 10 mg.

Complex sleep behaviours such as sleep-walking, sleep-driving, eating or making phone calls while asleep, with no memory of it: stop at once
Do not use
- Anyone with narcolepsy
Ask first
- You have liver impairment (moderate: maximum 5 mg; severe: not recommended)
- You take a CYP3A inhibitor or inducer (see the interactions list)
- You are pregnant (no human data)
- You are breastfeeding (no human data; it is present in rat milk; watch a breastfed infant for excessive sleepiness)
- You have moderate to severe sleep apnoea or COPD (not studied)
- You have depression or thoughts of self-harm
- You use another insomnia medicine or another medicine that slows the nervous system, or you drink alcohol
Watch for
- Next-day sleepiness: it can reduce daytime wakefulness, and the effects may last several days after stopping
- Driving was impaired in some people on 10 mg; the risk is higher with less than a full night left for sleep or a higher dose, so avoid driving and tasks that need full alertness then
- Falls, especially in older people
- Sleep paralysis, hallucinations at sleep-wake transitions, and cataplexy-like leg weakness
- Depression may worsen or thoughts of self-harm may appear: report any new unusual behaviour to a doctor at once
- Do not drink alcohol while using it

- Headache
- Abnormal dreams or nightmares
- Sleep paralysis (uncommon)
- Hypnagogic hallucination (uncommon)
- Complex sleep behaviour (frequency not known)
Medicines and vaccines used with it
- Itraconazole, clarithromycin, fluconazole, verapamil (strong or moderate CYP3A inhibitors): avoid
- Chlorzoxazone, ranitidine (weak CYP3A inhibitors): maximum 5 mg
- Rifampin, carbamazepine, St. John's wort, bosentan, efavirenz, etravirine, modafinil (strong or moderate CYP3A inducers): avoid, the effect is lower
- Alcohol: higher drug levels and worse balance and memory, so avoid it
- Benzodiazepines, opioids, tricyclic antidepressants and alcohol add to the effect on the nervous system; do not use with other insomnia medicines
- It lowers levels of bupropion and methadone (CYP2B6 substrates)

How is Dayvigo taken?
The package insert gives 5 mg no more than once per night, immediately before going to bed, with at least 7 hours left before the planned time of waking. A doctor may raise it to a maximum of 10 mg, depending on response and tolerability.
How long does Dayvigo take to work?
Blood levels peak about 1 to 3 hours after a dose, and about 2 hours later after a high-fat meal. Taken with or soon after a meal, it may take longer to fall asleep. The effective half-life of the 5 mg dose is 17 hours.
Is Dayvigo a benzodiazepine?
No. Dayvigo is an orexin receptor antagonist. It blocks orexin A and B, which is thought to suppress the drive to stay awake.
Is Dayvigo addictive or a controlled substance?
In the United States lemborexant is a Schedule IV controlled substance, and a study of abuse potential found it similar to zolpidem. In trials, no withdrawal effects were seen after stopping 5 mg or 10 mg.
Can I drive the day after taking Dayvigo?
It can cause sleepiness the next day, and driving was impaired in some people on 10 mg. The risk is higher with less than a full night left for sleep or a higher dose; in that case avoid driving and tasks that need full alertness.
Who must not take Dayvigo?
Anyone with narcolepsy. If you have liver impairment, take a medicine that affects CYP3A, are pregnant or breastfeeding, or drink alcohol, talk to a doctor first.
What are the side effects of Dayvigo?
The most common are sleepiness or fatigue, headache, and abnormal dreams or nightmares. Sleep paralysis and hallucinations when falling asleep are uncommon. If you do complex things in your sleep, such as sleep-walking or sleep-driving, stop at once.
